Brotherhood of the Silent Sorrow

This route will include the events that will allow you to progress in other Locations, then return to Brotherhood of the Silent Sorrow, after events, or encounters with NPCs. List of Equipment & Magic you can claim in this area:

Activate the first Prie Dieu.
Defeat the Warden of the Silent Sorrow
Interact with the locked door that requires a Key to the Chamber of the Eldest Brother.
Break the contraption on the upper right side of the door. This will take you to the next area: The Holy Line.

From Mercy Dreams

Buy the Quest Item: Key to the Chamber of the Eldest Brother.
Go inside the chamber and collect Relic: Blood Perpetuated in Sand from the treasure.

From Mountains of the Endless Dusk

Complete quests for Redento to help him cross.
Interact with the Mea Culpa Shrine at the left of the Prie Dieu that you have accessed prior to entering.
Look for the Jailed Ghost who will increase the slots of Rosary Beads by giving her a Knot of Rosary Rope.
Find Redento on another spot here with a boulder blocking his path heading to the right.
Head inside the door behind you. Navigate the path to the right until you reach the boulder blocking Redento's path.
Collect the Bead of Blue Wax next to the boulder. Afterward, you can return to Redento's spot easily.
Collect Tirana of the Celestial Bastion. This completes all the necessary items and NPCs in this area.

The Brotherhood of the Silent Sorrow is the game's starting area and serves as the game's tutorial section for various game mechanics. You can activate the first Prie Dieu which regenerates your health and Bile Flasks, or the container that The Penitent One drinks from to replenish his health. In the next room, you will fight Warden of the Silent Sorrow, your first boss fight. Once defeated, you will get the A Long Path Ahead achievement.

Continue exploring going to the left until you reach a locked door. You will need a Key to the Chamber of the Eldest Brother, this is a quest item that can be purchased from Candelaria, at Mercy Dreams. Leave this area for now, and break the contraption that's blocking by doing a jump attack. This will open up the path that will take you to the next area: The Holy Line.

Return to Brotherhood of the Silent Sorrow From Mercy Dreams

The Eldest Brother's chamber remains locked and you will only be able to open it by buying the key from Candelaria. You will find the first location of her shop in Mercy Dreams, where she sells 3 different items. You can buy it from her for 400 Tears of Atonement. Once you have the item needed, return to Brotherhood of the Silent Sorrow and unlock the chamber using the quest item. Go inside the room and retrieve the Relic: Blood Perpetuated in Sand from the treasure. This relic will reveal blood platforms across Cvstodia and will give access to areas that were previously inaccessible. This will help you explore rooms and areas further.

From Mountains of the Endless Dusk

Later on in the game, once you have completed quests for Redento to cross in Mountains of the Endless Dusk, you will gain a new entry point going to Brotherhood of the Silent Sorrow. There's a Prie Dieu prior to the entrance back to Brotherhood of the Silent Sorrow. Then, your next point of interest is the Mea Culpa Shrine at the left of the Prie Dieu that you have accessed prior to entering.

Climb the ledges going upwards and on the second level, you will find a Jailed Ghost who will increase the slots of Rosary Beads by giving her a Knot of Rosary Rope. You can do this for a total of 8 times. Continue exploring this area, and you will find Redento here again with a boulder blocking his path heading to the right. Instead, he will offer the door behind you. Inside this area, it will require expert navigation as there will be spikes that cause instadeath. The goal upon going inside here is to explore the right side, so you can break the boulder blocking Redento's path, and you will also find the Bead of Blue Wax here. Afterward, you can return to Redento's spot easily.

Your last stop here is to collect Tirana of the Celestial Bastion which can be found on the left side of the area after climbing the ladder. This will take you to a series of blood ledges that can be revealed by equipping Blood Perpetuated in Sand. This completes all the necessary items and NPCs in this area.
